To check the coil pack on a 5.3 Chevy engine for proper function without triggering a code, first inspect the wiring and connectors for any visible damage or corrosion. Use a multimeter to test the primary and secondary resistance of the coil pack, ensuring they are within the manufacturer’s specifications. Additionally, you can perform a spark test by removing the coil pack, reconnecting it to the harness, and grounding it while cranking the engine to see if it produces a strong spark. If all checks are good and no codes are present, the coil pack is likely functioning properly.

I found the easiest way to do this is read the codes, if it is reading a misfire on one cylinder, then swap that coil pack with one of the other cylinders coil packs, then you will have to clear the codes and drive it, then see if it throws a new code on a different cylinder.
