Depends on the type of car. Every new lock I have purchased has instruction with it for that particular model. Some are held in with a screw or pin. Some have a spring loaded stud that you depress with a paperclip or similar straight pin and it pulls out, it just depends on what you are working on.

To change the lock cylinder on a 1996 Saturn door lock, start by removing the door panel to access the lock mechanism. Then, detach the lock cylinder by removing the retaining clip or screw, and pull the old cylinder out. Insert the new lock cylinder into the housing, secure it with the clip or screw, and reassemble the door panel. Finally, test the new lock to ensure it operates smoothly.

To change the ignition lock cylinder in a 2001 Chevrolet Tracker, first disarm the air bag system. Then remove the clock spring assembly to be able to remove the old ignition lock cylinder and replace with a new.
If you are talking about the ignition lock cylinder, the steering wheel must be removed and the column disassembled to access a lock screw which holds the ignition lock cylinder in the column. No key is required for this (unlike all other vehicle manufactures). If you are talking about the door lock cylinder, the door panel must be removed, and the lock cylinder retaining clip must be removed (again no key is needed, which is the same for all manufacturers). If you are talking about the trunk lock cylinder, it is easily accessible from inside the trunk. Those are the only types of lock cylinders in this type of vehicle. To replace lock cylinder in the hatch ,first inside hatch panel must be removed. After removing the panel you will have access to the lock cylinder. Remove the linkage to the cylinder and pull the retaining clip from the cylinder and the lock can be removed from the outside of the latch. This is not a hard job to do , 1 hour max if your slow .
To rekey a lock and change the key that operates it, you need to remove the lock cylinder from the door, take out the old pins inside the cylinder, and replace them with new pins that match the cuts of the new key. This process allows the lock to be operated by the new key instead of the old one.
