The gearbox found in a car allows the engine to perform across its most efficient rev range - or across the range it produces the most torque.
The S is for sport and then 1st gear. When you swap over to S your gear changing will alter and will take longer to change gears allowing you to go higher up the rev range.
The flashing is indicating that your gears are disengaged and ready to shift. Brake must be depressed.and obviously vehicle fully stopped. The beep.and flash will cease when fully and successfully shift, signalling vehicle ready to drive.
