This is one part of a two part answer.
Q: Does each discrete colony represent the growth of one cell? Explain your answer. Why can a single colony on a plate be used to start a pure culture?
A: No because a discrete colony is the growth of the descendants of a single cell. The cell will self replicate it self on the growth medium to produce more copies of it self thus creating a colony. A single colony has many cells in the colony. Cells from the colony can be extracted and then placed in a pure culture for better growth.

Epiphyseal plate
The thin band of cartilage where bone growth occurs is called the epiphyseal plate. This plate is responsible for lengthening bones during the growth phase in children and adolescents. Once growth is complete, the plate ossifies and is replaced by solid bone.
Sources of error in viable plate counting include inaccurate dilutions leading to over or underestimation of colony forming units, uneven distribution of bacteria on the agar plate causing inaccurate colony counts, contamination from environmental sources impacting the results, and variability in the incubation conditions affecting bacterial growth rates.
The growth plate of a long bone is located at the end of the bone, near the joint. It is also known as the epiphyseal plate and is responsible for longitudinal growth during childhood and adolescence.
The growth plate, also known as the epiphyseal plate, allows the diaphysis of the bone to increase in length until early childhood. During growth, the cartilage in the growth plate is replaced by bone. The articular surfaces are shaped by the action of osteoblasts and osteoclasts during development and growth.
it hurts quite bad. I fractured my growth plate in my shoulder.I took a couple x rays and it had a tiny crack in the growth plate.

The following are some advantages of an agar plate verses a slant tube: 1. Surface area- An agar plate has a much larger surface area: a. Easier to isolate individual colonies using the streak-plate method. i. Evaluate the colony shape, margin and elevation. b. Can grow a larger number of cells. 2. Growth- An agar plate allows you to quantify the number of colonies on an agar plate, provided it is within the 30-300 range. Whereas the slant tube cannot quantify growth but only describes growth as none, slight, moderate, or large.
In those who are not fully adult, growth plates are located on the ends of long bones. On the arm, the growth plate is located on the end of the humerus near the elbow.
When the epiphyseal plate is replaced by bone, then growth at that bone stops.

Advantages: 1. Counts only living cells 2. Standardized test - used worldwide Disadvantages: 1. 1-2 days of incubation 2. Melted, heated agar 3. Osmotic shock
