Many microscopes these days are capable of "live imaging". Viable cells can be placed under the microscope, the lens magnifies the image, and the magnified image can be sent to a highly sensitive digital camera. You would probably need to use some stain, something that binds DNA, to make the chromosomes easily visible within the tissue.

What stage is the mitotic spindle visible?

Chromosomes are easiest to see when they are condensed(coiled and supercoiled, so that they become short and fat). They are at their most condensed from late prophase through to early telophase, in either mitosis or meiosis.Their visibility also depends on suitable microscopic technique, including the use of an appropriate stain, such as acetocarmine.
