Lipid and steriod synthesis will not work properly, toxins will not be detoxified properly, and there may be problems with intracellular calcium signaling, which can be very disruptive to cellular process as calcium is a very import signaling molecule in cells.
If the ER stopped working, then the cell wouldn't have proteins, and sequentially die
If the Rough Endoplasmic Reticulum is absent, the cell explodes. The ER is absent in the red blood cells of mammals.

If your nerves stopped working you could not move, because your spinal cord helps your body to move even in the the smallest parts of your body.
the earth will become extinct eventually
Continual spasms until the nerve is repaired, which takes years and years if ever.
your body wouldn't be able to grow, regulate blood pressure, regulate water balance, or activate other glands.
If an organelle fails then the cell will die. I am assuming you mean if say the mitochondria as a whole stopped working, or if the endoplasmic reticulum stopped working in the cell as a whole. If an organelle system died in every cell, the entire organism would probably also die or not work correctly.
smooth endoplasmic reticulum
There will be no distribution and storage of enzymes in the cell.
Synthesis of proteins occurs in the Rough Endoplasmic Reticulum.
You will die.

If it stopped working then you would die. You wouldn't die if you were put on the a machine right away.
