to access your flash drive you should be able to open my computer and then listed with the drives should be your flash drive you can just right click and then go to open. However, if this does not work do the following to convert to FAT or NTFS:
To convert a volume to NTFS or FAT from the command prompt

In the command prompt window, type
convert drive_letter: /fs:ntfs
For example, typing convert D: /fs:ntfs would format drive D: with the ntfs format.

Windows Vista does not officially support booting from a FAT32 drive, although it is technically possible. Vista supports FAT32 on Flash drives and hard drives, although it will not format a hard drive over 32 GB as FAT32.
Your best bet will actually be to use a USB Flash Drive with folders named exactly like those found on the PS3's internal hard drive. It has to have a FAT32 file system, as well, so if you use an external hard drive you'll have to make a partition. Note that FAT32 only allows up to 4GB for one file.
