This depends on what actually causes it to be defective.
The easiest and cheapest in many cases would be to buy a universal remote control.
If you really need to fix it, then I would suggest checking that batteries are giving power to the circuitry inside. Often with a battery leak, the wires inside get heavily corroded and the remote control might be faulty due to this.
Buttons are a place of wear and tear. Often not so much wear, just a bit of dust here and there that might cause buttons to be hard to push.
A soft moist cloth with a little bit of soap and spirits takes care of this problem. I always find small cotton swabs quite useful.
Some times, the wear and tear of a remote control is so that the actual IR Led's themselves are faulty. Try to find some information on internet regarding your remote control. Try to find out what wavelength the diodes emit light on. When you know that, these diodes can easily be replaced by new ones. One can of course just buy a few different ones. The chances are that they will work even if they are not a perfect match. The way to tell if the diodes are working is to look thru your camera phone or any other electronic camera at the remote, push any button and you should see the lamp light up even though you can't see the light with just your eyes.
Remote control is a wide subject. So far written Mainly for those remote controllers for TV/Radio/stereo/and similar.
