It means it's your turn to buy a round of drinks. Australian slang. "It's my shout" = It's my turn to buy a round of drinks. Presumably because one will shout to get the bartender's attention. If someone says "Your shout, mate" head immediately to the bar and buy a round or a pitcher of beer.
