No. It is concentrated, but urea is considered generally non-toxic, needing extremeley large amounts to cause damage. Far more than a hamster can produce.

You can tell if something is wrong with your hamster if it starts acting different to how it normally does, waking up later, not coming out for food, being irritable etc. Also when hamsters become ill, many lose their hygiene of going to the toilet properly, their tails may be sodden with wee.
