Series creator Naoko Takeuchi has said in interviews that only girls/women can be Sailor Scouts [Senshi in the original Japanese], and that she herself was surprised when the animators of the fifth and final season chose to make the Sailor Starlights gender-changing Senshi, that start out as males and become females when they transform.
However, it was implied in the anime that the male gender that Taiki Kou, Seiya Kou, and Yaten Kou each possessed as humans was in fact just a "disguised," based off the comment that Princess Kakyuu made upon meeting them once more.
There is also no problem with a Senshi cross dressing, which is what the Sailor Starlights did in the original manga (no one recognized them as females, even though that's what they were, all the time), and as Amara [Haruka] Ten'ou (Sailor Uranus) did while infiltrating Infinity Academy in season 3 (Sailor Moon Super).
It has been said that Darien [Mamoru] possesses the Sailor Crystal of the Earth, but he doesn't transform the way the Sailor Senshi do, though he does "transform" into Tuxedo Mask and Prince Endymion.

No, in series one they die but the power of the Silver Crystal brought them back to life, and they can't permanently die as in the future Rini knows them and gives Serena messages from them in the future Raye (calling her stupid). ---- However in the last series the Senshi (Scouts) and Mamoru (Darien) all die but Sailor Moon with the help of Chibi Chibi brought them back to life once Galaxia had been defeted.

Sailor moon doesn't ever die in any of the episodes. In episode 47, i think, Queen Serenity as a hologram visits the Sailor Senshi (the scouts) And tells Sailor Moon about her past life as Princess Serenity. Queen Beryl takes Prince Endymion of the earth a little bit before Princess Serenity jumps up to Endymion, thus leading to Beryl firing and killing both Endymion and Serenity. So, Sailor Moon really does die (but in her past life)
