Yes. Once Legendary Six Samurai Shi En is successfully summoned to the field, then it can use its quick effect against spells or traps the opponent activates, even one activated in response to its own summon.

∙ 10y agoYes, Shi En can negate a Spell or Trap that activates in the graveyard. The only difference than normal is a 'destroy' effect does nothing to a card in the graveyard, or in this case, the banish zone, where Breakthrough Skill will have sent itself as its own cost.
