The Joker would be known by many comic fans who are also SF fans, but many Star Trek fans would only have peripheral knowledge of the Joker or other Batman characters. As both Spock and the Joker have appeared in several high-profile movies, they would both be known to many non-fans.
Spock, however, is something of a cultural icon in the US, and despite having been around for 3 decades less than the Joker, would probably be considered more historically famous.

Spock is way more famous than Harry Potter ever though of being because Spock has been a much more around and is a much more fascinating character.
