It was a combination of factors. The fort was built on swampy land so mosquitoes were very bad and they carried diseases. The water supply was bad and that added to disease. The men who landed there would rather look for ways to get rich rather than start a settlement so the work that was needed to be done didn't get done. The Native Americans in the area were not happy with the settlers and a few were killed by them. There was even a Spanish spy in the group. Put all of these things together and you get dead people.
