They're all designed for the same purpose - to kill something. Although smaller is not necessarily less lethal, smaller calibers generally will be designed for short-range use on small animals and large calibers may be suitable for hunting bear.
Dirty Harry's line about the 44 Magnum being the most "powerful handgun in the world" has not been true for some time. There are many handguns chambered in rifle calibers for hunting and long range shooting. These pistols, in calibers such as .308, 8mm, 30-06, etc, are far more powerful than the .44 Magnum. In recent years, large bore revolvers have been made in very powerful calibers, such as the .460, 454 Casull, .500 S&W, etc. There are even a few special handguns that fire the 12 g shotgun shell. However, a challenger for the title of the "world's most powerful handgun" may be the Maadi bolt action pistol, in caliber .50 Browning Machine Gun. Extremely heavy, large, expensive, and of limited usefulness- but powerful. There is also a one of a kind custom pistol in .600 Nitro Express. There are videos online of a man shooting it where the recoil rips it right out of his and and flings it behind him.
Depends on how much money you have. Springfield Armory makes an excellent and very safe pistol called the XD. Available in 9mm, .357Sig, .40 and .45 calibers. Your best bet is to go to a local gun shop that has a range that rents guns. Try out different models and calibers until you find one you like. Join a local gun club and ask the members questions.
Both the BAR (Browning Automatic Rifle) and the Browning machineguns (series 1919) were made in caliber 30-06. HOWEVER, there were other calibers manufactured for special markets. Search Wikipedia for BAR, and look up some of the secondary calibers- they may be a surprise.

It's difficult to answer what is "average." Pistol bullets can be as small as .17 caliber (although not common) and as large as .950 caliber (also very uncommon). The most common semi-auto handgun calibers are .22, 9mm, .40 (depending on where you are) and .45. The most common revolver calibers are .22, 38, 357, and .44, but there are a lot of other calibers available.
