At the courthouse town of Appomattox, Virginia. The formal surrender took place in the parlor of Wilmer McLean. Interestingly, First Battle of Bull Run was fought on property owned by him. McLean moved to Appomattox in the spring of 1863 to escape the seat of war in Northern Virginia.
Additionally, for all practical purposes this ended the war, however, there were other armies and naval vessels that surrendered later in April and even far into 1865.
It essentially brought the Civil War to an end.

Generally speaking the surrender of Robert E. Lee to US General Grant is referred to as the surrender at the Appomattox Court House. Lee's surrender was the key which in all practical terms ended the US Civil War. Later other Southern armies would surrender.

Well, when Cornwallis surrendered, it had been just a battle that he lost. But when Robert E. Lee surrendered it was the whole reason to why the confederates lost the Civil War.
