The beginning of the end occurred with the assassination of Alexander Severus in 235 AD, after which a succession of short term rulers failed to establish control. Ultimately, a number of failed invasions from foreign armies weakened the power structure of the empire until it eventually met a gradual division and Ultimate demise with the end of the Caran Dynasty in 285 AD.
There are no easy answers to this question, in large part because the empire was divided into two separate entities in the West and the East. The Western Empire declined gradually, but faster, than the empire in the East. The Western Empire officially came to an end when Odoacer forced the last Western Roman Emperor, Romulus Augustus, to abdicate in 476 AD. This event, however, was presaged by numerous occasions which demonstrated that the Western Empire, for all intents and purposes, had ceased to exist. The Eastern Empire suffered a much more slow and gradual decline, and survived until the Ottoman Turks captured the capital at Constantinople in 1453 AD.

The two halves of the Roman Empire split before it fell. The end of the empire in the west is referred to as the Fall of Rome and happened in 410 AD. The Eastern half became the Byzantine Empire and lasted another thousand years.
The West was subjected to massive inflation, a drop in the value of money, as string of terrible emperors, a sharp drop in the effectiveness and loyalty of the army due to the number of mercenaries and the Germanic tribes on its borders. The muslim Ottomans ate away at the borders of the Byzantine Empire until the capital of Constantinople was all that was left. And in 1453, the walls of the city fell and that was the final end of all of Rome.

Historians give 476 and a date for the end of this part of the empire.
The eastern part of the empire was not affected by the Germanic invasions and continued to exist. Later it, too, lost many lands due to invasions by Arabs and Slavs, but it managed to survive in a reduced version until 1453 when the ottoman Turks took over its capital, Constantinople.
