Assuming the Duke and Duchess of Cambridge's first-born child is male he will be a prince, and will be 3rd in line to the throne, after Prince Charles, the heir apparant, and Prince William, the heir presumptive. However, under the proposed changes to the Act of Succession agreed last October at a Commonwealth Summit in Perth, Australia, but which have still to be ratified by allthe 16 nations of which the Monarch is Head of State, the primogeniture principle will in future decide the future monarch. Thus, if William and Catherine produce a daughter first, she will be heir presumptive, even if their subsequent offspring are male.

Yes, any child he has will be placed on the line of succession, after himself but before his brother Harry. As the system is biased towards males, it means that a younger son of his will be placed closer to the throne than an older daughter, but the daughter will always be higher than William's own brother.
