It's called a mint mark and identifies the facility where the coin was minted. Modern circulation coins use P for Philadelphia and D for Denver.

Proof coins may have an S for San Francisco and bullion coins may have a W for West Point.

Other mint marks have been used in the past. Tracking them all can become quite complicated.
