The Missouri Compromise of 1850 affected Kansas & Nebraska territories. The Compromise was an extension of the one made in 1820. The idea behind the compromises was to maintain the balance of free states and slave states. Now the statehood of Kansas & Nebraska came into the mix. The consensus of Congress was that left to themselves, the people of Kansas would follow the lead of Missouri and vote to become a slave state. Also, that the people of Nebraska would vote for a "free state". Later events, such as a major conflict in Kansas & the soon to come Civil War would send the compromises into a free fall.

The Missouri Compromise was Missouri (South) and Maine (North) wanting to come into the United States union, government didn't know if they both should be slave states or free states. An idea proposed by Henry Clay stated that Maine should be a free state, so Missouri should be a slave state. This achieved that any new state that wanted to become apart of the union above the 36'30 latitude are free states, then anything below the latitude are slave states.

This was prior to the civil war ( 11 years later the civil war will start) so much of what happened was about slavery. In 1846 Dred Scott sued for his freedom in a Missouri Court and in 1852 the Missouri Supreme Court ruled against him. On March 1857 Chief Justice Taney of the United States Supreme Court against Scott. He wrote that slaves were property. In 1853 the Kansas Nebraska act was passed and in 1855 Bleeding Kansas happened when the vote came to decide if Kansas would come in as slave or free state. On May 21, 1855 John Brown and his sons attack a settlement near Pottawatomie Creek and murdered 5 people. This won't be the last time John Brown is heard from. So, as you can see it wasn't one event but many in the 1850's that were important. Each of these will help lead the country towards civil war.
