There is no transmission drain plug. Your transmission uses the same oil from the primary. Draining your primary drains your transmission.
That plug is the drain plug for the primary and transmission oil supply.
The oil drain plug is at the bottom of the oil tank (for engine). Bottom rear of primary case for primary & transmission.

The transmission drain plug is located on the left side of the bike as you are sitting on it, right behind the front foot peg. It is a 16mm hex head bolt.
To change the transmission fluid in a 2006 Harley Sportster 1200, first ensure the bike is on a level surface and warm the engine slightly. Remove the transmission fluid drain plug located at the bottom of the transmission and let the old fluid drain completely. Replace the drain plug and refill the transmission with fresh fluid through the fill plug on the side, using the recommended type of transmission fluid. Finally, check the fluid level to ensure it's at the correct height and secure the fill plug.
