Yes, Zira loves Scar. She loved him enough to make it her life's purpose to train his chosen heir, Kovu, to follow in his pawprints, and become the king by killing Simba. However, their relationship is a "leader-follower relationship" in which she admires and respects Scar as a leader, and blindly lives by his values and ideals. The two are not mates because Zira's children's (mostly Kovu) relationship with Scar was changed to avoid an incestuous relationship with Kiara during production of The Lion King II.

However, sometime before the events of The Lion King II, Zira is known to have had some relationship to Scar although it is has not been officially known, but it is implied that their relationship could have been romantic. In a deleted scene from the movie, Zira says that if Scar were still alive and still king, she would be his "loving, devoted queen" and had cubs such as Nuka, although it was not confirmed that he is Scar's blood-related son. So, it is unknown whether the cut scene still applies to the movie as it was deleted from the film. So, there is no official knowledge if Zira is related to Scar.
