Captain America is the first to actually join the team. "Iron Man" was the first of the movies that lead up to "Marvel's The Avengers". He was the first one that Nick Fury asked to join the team but after seeing Natasha Romanoff's report he changed his mind and decided he doesn't qualify.
And since Fury didn't even ask any of the others in their respective movies, Captain America is the first who is officially on board.
