The Great Rift Valley was caused by the edges of two tectonic plates, not by a river.

There are rivers in it, though, notably the River Jordan, in the north, which flows into the Sea of Galilee and then into the Dead Sea.

The Western Rift, in East Africa, contains some of the deepest freshwater lakes in the world, but no outlet to the sea.
