You might actually look at some of the Russian Space Programs as they have had more experience with long term space exposure. One of the notes I have seen is the recommendation for rigorous exercise programs of approximately 2 hrs/day to prevent decalcification of the bones and weakening of the muscles.
There certainly has been significant discussion about adapting environments in space to the human needs, for example giving some semblance of gravity.
