Many things. First and foremost is the digging they do in their backyard in the dead of night. Tom Hanks (Ray Peterson) character sees this himself, as does his son.
Second is the fact that they take out their trash in the dead of night. Not only that, but they drive the garbage down to the curb, because apparently it's too heavy to carry. The neighbors become suspicious of what's in the trash bags: they seem squishy yet solid, like flesh. But it's pouring rain at the time, so they decide to come back early in the morning to open up the bags. But when they do, the mysterious trash bags are gone.
Furthermore, they never seem to go in and out of their house, except in the middle of the night, nor do they ever have any visitors or deliveries. In the words of Ray's buddy and neighbor, Art Weingartner, "What do you think they're eating over there?"
How they got the house, and why they moved from their old house, is also a mystery. Their old house apparently burned to the ground mysteriously, and no one saw any moving vans or trucks when they moved into the new house. Nor did anyone see the last residents of the house move out.
Their general unfriendliness, as well as the fact that their house and yard are a shambles compared to the neatly kept lawns and homes of their neighbors, is also a factor. They turn out to be even weirder and unfriendlier in person, when some of the neighbors on the block go over to their home to say hello and to check them out. For example, one of them, "the doctor," has what seems like blood on his hands when he comes up from the basement.
Also, they have bars on their basement windows, and on one occasion, the neighbors see a fantastically bright electrical blowout coming from their basement. They also have extremely elaborate burglar alarms set up.
There's also the fact that their elderly neighbor, Walter, disappears mysteriously, and Ray finds his toupee in their home. However, it turns out that Walter had a heart attack and was in the hospital, and the reason the toupee was in the Klopek's house is never explained.
Oh, and Ray's dog digs up a femur (a human thighbone) in the Klopek's backyard.
