Zeref was obsessed with researching the nature of life, death and magic, which is where the designs for the R-system (Tower of Heaven arc) and Eclipse came from. He did this as he wanted to resurrect his younger brother, who had been killed. He was warned not to continue this research as it was against the sacred teachings of the god Ankhseram, but Zeref did continue. He was duly cursed with the 'Curse of Contradiction' - the more Zeref values life, the more lethal he becomes to people around him, but when he 'forgets' the value of life, the curse can be controlled. The curse also made him immortal.

Zeref had simply withdrawn himself from the world, going into seclusion on Fairy Tail's holy island of Tenrou Island. He claimed he had become tired of his life of evil, and wished only to die. He said that he had 'learned the value of life', which had the side effect of letting his death magic powers run out of control, hence his need to hide. However Grimoire Heart's interference angered him. He said people like them make him forget the meaning of life, regaining his full powers. In the 'Fairy Tail Zero' story we learn that he befriended Fairy Tail's founder Mavis in the past, and taught her some of her spells. Her usage of an incomplete form of the 'Law' spell caused her to be affected by his curse of immortality. After she accidentally caused the death of 'Rita', Makarov's mother, she too withdrew herself from others, to avoid doing the same. She met with Zeref, wondering if they could live together happily without the risk of harming each other. Unfortunately Zeref's curse was stronger, and put Mavis into a coma. Later Zeref is revealed to be Emperor Spriggan, ruler of the mighty Alvarez Empire. He has declared war against Fairy Tail, with the ultimate goal of capturing 'Lumen Histoire', the crystal containing the body of Mavis in a form of magical stasis. This would grant him nearly unlimited power, enough power to defeat the dragon of the Apocalypse, Acnologia. Zeref fears the dragon, because he has the power to wipe out mankind. Zeref cannot die, so would then face being Acnologia's plaything for all time. After Mavis was released from the crystal, Zeref successfully extracted her powers and became 'white mage' Zeref. Even these extraordinary powers were not enough against Natsu though, and he was defeated in a climactic battle. As he lay there, recovering, Mavis approached him and the pair reconciled. With their feelings for each other now balanced, they successfully killed each other with their curses, bringing the story of Zeref to a close. In the final issue, we see that he and Mavis have been reincarnated as new people with new names, fully mortal, who have a chance meeting and quickly become friends. That's the last we see of them.
Why in Fairy Tail is Zeref with the Natsu group in the opening for episode 221?
Zeref doesn't appear in 'Break Out', the intro for episode 221. At a guess the character is actually Rogue Cheney who appears with Sting and Team Natsu near the end of the intro sequence.

How does fairy tail end?
Black Mage Zeref launches an attack on Magnolia. He captures Fairy Tail's first guildmaster Mavis (who is revealed to be still alive, sealed in a crystal) and steals her power. Becoming White Mage Zeref, he deals Natsu a mortal wound before preparing to rewind time to relive his own life at the cost of undoing 400 years of reality. Meanwhile Lucy is in possession of the Book of END, the book which Zeref created to revive his brother Natsu 400 years ago. Seeing the words disintegrate due to the wound, she rewrites the missing symbols to heal him. Natsu rises and is able to strike Zeref down, burning the stolen magic away, though Zeref does not die due to his immortality. Mavis approaches him, declaring both her love and hatred for him. Realising that their twin curses are the key to ending their own immortal lives, they kiss, and move on. Lucy finishes rewriting the book, so when it vanishes due to Zeref's death, Natsu survives. Their victory is cut short as the other antagonist Acnologia survives a trap to delete him from time. He swallows the seven dragon slayers into a private realm as a source of energy, while his body goes on a rampage. Inside, Natsu and the others fight Acnologia's spiritual form, while the mages think of a plan to stop his dragon body. Eventually the mages trap him inside Fairy Sphere, which paralyzes his spiritual side too and allows Natsu to strike a deathblow. The guild is rebuilt and life goes on as normal - Zeref and Mavis have been reincarnated as different people, though meet and take an instant liking to each other.
