The opening shots were at Fort Sumter, a Union garrison on a tiny island in Charleston Harbour, when Confederate artillery opened up on it. The garrison was evacuated without casualties, except one by accident.

But this does not really qualify as a battle, nor do the small skirmishes in Virginia that preceded the first pitched battle of the war, Bull Run (Manassas), July 21st 1861 - a Confederate win.
