An overwound pickup (or humbucker) is a term that is used for pickups that receive more wounds to their coils, hence making them hotter.
The more wounds a coil has the louder the pickups get. Usually, people refer to this as being "hot". While the pickups get louder with more wounds, the resonent peak of the pickups is also lowered. The result is usually that a hot pickup is darker, and a low output one a bit brighter.

On an electric guitar, a middle-position humbucker can, but will not necessarily, interfere with a player's picking or strumming movements. The primary consideration is the location (especially the height) of the pick-up itself. Also to be taken into account is the typical positioning of the player's picking/strumming hand.
Its purely a matter of choice, the humbucker pick up will give the Stratocaster a different sound.

Yes it is possible to do so. There are a few ways of achieving this, the easiest way being to buy a new pickguard routed for humbuckers (either Humbucker-Humbucker or Humbucker-Single-Humbucker). A good place a new pickguard to get on is on guitarfetish.com. An other way is to route your current pickguard, but do not recommend this as it is much more risky. I also recommend getting the wireing done by a professional if you do not have experience in the domain. Nevertheless, the wireing diagrams can be found on seymourduncan.com .
