As of this post 8 Aug, 2012, at a minimum, there are over 120,000 songs using Vocaloid.
On the Japanese website, Nico Nico Douga (the central hub of Vocaloid music and videos), we get these numbers:
1. 84,000+ original songs tagged with "Vocaloid" and "オリジナル曲" (original_song).
2. 47,000+ cover songs tagged with "Vocaloid" and "カバー曲" (cover_song).
Take the above 120,000+ songs from that website alone and consider the following:
1. Add all the potential Nico Nico Douga videos that were tagged incorrectly.
2. Add any original songs and covers from YouTube that are not reprints of existing videos.
3. All other websites that have original music and covers using Vocaloid.
The number is possibly much higher than 120,000.
The Vocaloids were designed without any relationship backstories. However, there are a lot of fans that pair Len with Miku. There are also many fanmade Vocaloid songs that feature Len having feelings for Miku.
Of the four 'official' Vocaloids. I think Miku Hatsune is the most popular. I didn't go around the world in a plane counting Vocaloid fans, and I didn't look this up on Google, or Bing, or whatever. But, Miku has the Most songs. I might not be the most reliable soarce, so... I would look up songs by different Vocaloids on youtube and compare the views and likes. No. VOCALOIDs are not couples. VOCALOIDs do not have to deal with anime or couples. It's only a singing program.

vocaloids isn't a anime or a tv show, their voice synthesizers. You can find PV's about their songs on youtube
you can find most of it on youtube, and some on specialty sites - but most animes have no official english versions of their songs. most will be fan-made or vocaloids.

There is no any other VOCALOIDs. All vocals found in VOCALOID wikia are the original VOCALOIDs made buy actual music/recording companies. Haku Yowane, Akita Neru, and Hatsune Mikuo etc. are fanmade based of the VOCALOIDs voicebanks.
