To get into Bartleby, you need to defeat ALL of Wizard City. Meaning Lord Nightshade, the final boss. After that Merle Ambrose will tell you to go to Krokotobia. You will then need to use Bartleby to get there. Before that there is no other interaction with the tree itself, exept maybe for a quest or two when you need to talk to him.
