in tissue culture should maintain some steps mandatory example explanting, surface sterilization, inoculation , incubation and subculture so their is no any simple protocol for tissue culture especially in banana need to take care still more than others plants because endogenous microbial contamination's will be more in succors so its better to follow specific protocol that should give healty plantlets free from diseases instead of going simple one.

How does a banana reproduce?

Banana plants have flowers, and wild bananas reproduce just like other flowering plants. So insects will go to the flower to collect the nectar, and the pollen will stick to them. The pollen will then drop off at another banana plant thus reproducing the banana plant. Commercially cultivated bananas are sterile and are reproduced, or propagated, by suckers or by tissue culture. See link below for more information.
