Mercutio is spoiling for a fight with Tybalt. When Tybalt asks if he can have a word with Mercutio, Mercutio says "Why not couple it with something? Why not make it a word and a blow?" He is asking Tybalt to slap him, to challenge him to a fight. Tybalt says he would be happy to, but right now he has other fish to fry.
Tybalt was enraged that Romeo was at the party and he wanted a sword to kill him. However, Capulet told him to leave Romeo alone because he was not causing any trouble and he had heard good things about Romeo and he did not want to be rude to his guests.
He gets angry, very angry! He knows that Romeo is a Montague and assumes that the only reason why a Montague would show up would be to "fleer and scorn" at the Capulets. One thing: Tybalt has a short temper/easily angered, which might have been the reason for his hate, even though Romeo refused to fight Tybalt.
Kill him and serves him right for gatecrashing the party.
