Romeo is playing with words. He says that if his holding Juliet's hand (which he calls a holy shrine) is profane, or irreligious, he will make up for it because his lips are pilgrims (holy people) who will "smooth" the problem with a kiss. In this way he hopes not only to be forgiven for holding her hand without permission but also to get a kiss out of it. The "pilgrims" are "blushing" because of the ruddiness of the lips.
