If Iago had told Othello right out that they were having an affair, Othello might consider that he had a motive for saying so. It would become an issue of "Who is lying? Iago or Desdemona?" and Othello would probably believe Desdemona. But by planting the seed of suspicion and then feeding it by downplaying it, Iago makes sure that Othello is not even aware that it is Iago that is telling him about the supposed affair. He thinks he's finding it out for himself. This way it does not become a credibility issue between Desdemona and Iago.
Roderigo wants Desdemona as his wife, which makes Othello his rival. It is natural for people to be hostile to their rivals in love. As for Brabantio, Othello is a parvenu, not an established member of the Venetian society. And he has married Brabantio's daughter without first discussing it with the father of the bride. In the society presented here, as in many nowadays, a father felt he had rights of ownership over his daughters, which would make Othello (like Romeo in Romeo and Juliet and Lorenzo in Merchant of Venice) a thief.
We hear about Desdemona in the first scene when Iago tells Brabantio that she has eloped with Othello. She makes her first entrance in Act 1 Scene 3 and her first line is "My lord, I do perceive here a divided duty."
Othello is a General that lives in Italy(Venice), but is a Moor. Traditionally that means he is black while everyone else is white. He is very respected and marries on of the leading families daughters. Iago is one of his most trusted lieutenants, but after great victories Othello decides to promote someone else. Iago then begins a scheme to get power. First he brings down his other rivals for power. Then he seeks revenge on Othello. To do so he starts scandals almost on a whim and makes everyone distrust one another. Including Othello and his wife. Side note: Othello has epilepsy and no one must know or they will think him weak or possesed. Iago forces Othello to smother his wife and all of his rivals to die, yet in the end He loses his own life in his web of lies. One of the most Ironic things about Othello is Iago the most wicked liar coins the term (I wear my heart upon my sleeve.) Meaning he does not decieve. Iago makes the top ten most evil fictional villains ever.
More than anything, his humility. Humility is a good quality, but Iago makes Othello's lack of pride work against him. When it comes to fighting in a battle or dealing with a load of hoodlums in the street, Othello is all confidence: he knows how to deal with those things and he knows he knows it. However, in matters of love, he is not so sure. He has reached middle age without really having to deal with women. His relationship with Desdemona starts off with him telling her stories of his life. He isn't trying to woo her, to pick her up or in any way get her to love him. But she does love him. Othello is amazed that she should fall for someone who is old, who is not of her class, who is a foreigner and has an unpopular skin colour. Why should this wonderful young woman pick him of all people? We can see through Desdemona's eyes past these superficial qualities to the qualities of the man within. Othello is competent, commanding and very human. His very humility makes him a good commander and leader. It also makes him doubt whether a woman like Desdemona could love someone like him, a doubt which Iago exploits and turns to the certainty that she must love someone who appears to be more deserving--someone like Cassio. In Othello more than any Shakespeare play we see the pointlessness of the "tragic flaw" type of analysis, which tries to make the plays into parables designed to convey trite bits of morality. The characteristic which leads to Othello's demise is a virtue, but as those compassionate and generous souls who have been conned by phoney charities know, even virtues can be exploited by the wicked. Both Othello and Desdemona are very good people, and yet their doom overtakes them anyway, and in part because they are so good. That's what makes it so heart-wrenching.

Iago opposes Othello because Othello chose to promote Cassio to lieutenant instead of Iago. Also, Iago is prejudice against Othello for his skin colour. Othello was a victim of the time he lived in.
There is in effect a trial. However, Othello's humble, straightforward and reasonable reply, backed up with Desdemona's level-headed and balanced evidence makes Brabantio look a fool with his frenzied rants about witchcraft and the like.
