Lady M: I heard the owl screech and the crickets cry.
Didst thou not speak?
Macbeth: When?
Lady M: Now.

Macbeth: Hark! Who lies in the second chamber?
Lady M: Donalbain.
The Macbeths are jittery about whether someone might wake up and catch them in gore up to their elbows. Macbeth thinks he heard a noise, and Lady M thinks she heard someone speak. Macbeth thinks that he has heard it again--he says "Hark" which means "listen". He is asking Lady M to listen for the noise he hears. Then he wonders whether it might have come from the second chamber (next to the king's), and asks who is sleeping there. For some reason, knowing that it is Donalbain satisfies him and he can be distracted by the blood on his hands.
