It is essential in this part of the plot for an obstacle to arise between the lovers. That's how romance plots develop: the couple falls in love, but something happens to keep them apart. In a romance with a happy ending, the obstacle is overcome and the couple united or re-united at the end. Not in this romance though.
The obstacle which Shakespeare raises is a legal one--it becomes illegal for Romeo to live in the same city as Juliet does. Only a very serious crime can bring this about, and Tybalt has been built up as an antagonist, someone who is determined to fight with and kill Romeo. But at this stage Romeo is not in any way hostile to the Capulets who are now his relations. He is not likely to agree to a fight. His anger overwhelms him. What would make him that angry at Tybalt? There are of course lots of possibilities, but Shakespeare has Tybalt kill one of Romeo's friends. Tybalt might have killed Benvolio but Benvolio would have refused the fight (as he tries to stop the fight between Tybalt and Mercutio). If Tybalt killed him it would be murder, and although it would have made Romeo mad, it would not have made him as mad because Romeo could not have thought that he was At Fault.
This is just guesswork, but it is a plausible estimation of how Shakespeare might have decided that both Tybalt and Mercutio must die.
