Yes, they did. In fact, the Republicans have set a record for filibustering (and thus blocking) legislation the president has proposed. I enclose an article about the number of times the Republicans have blocked an up-or-down vote on proposed legislation. It should be noted that the link is considered an opinion piece, but the authors are two respected political scientists, one of whom is a conservative and one of whom is a liberal/moderate.

It is difficult to tell, since the 2012 election has not occurred yet. It should be noted that some very prominent Republicans, including Gen. Colin Powell and Col. Lawrence Wilkerson, have come out in support of the president, and there is also an advocacy group called Republicans for Obama; these are current Republicans who believe the party has become too extreme and wish the moderate strand of Republicanism would return. Until it does, they say, they will work for moderates like President Obama. I enclose the link to their site. It should also be noted that there are some Democrats who are supporting Mr. Romney. In any election, there are people who may leave their own party (temporarily or permanently) because the nominee of the other party more closely shares their views. There are politicians on both sides who have done this, and in the 2012 election, we are seeing some examples too.
