Spring Street is a couple blocks south of Houston, and the Houston Street Station on the 1 train is actually on Varick Street between King and Houston Streets.
It's pretty simple. Take the Downtown 2 or 3 (of the 1-2-3, the red line) from 34th Street-Penn Station to Fulton Street (at William Street). Then walk east on Fulton Street until you get to the East River. You will know you're walking east because the next street you'll reach will be Gold Street. If you walk one block on Fulton Street and end up on Nassau Street, then you have walked west, not east.
Take the Uptown E train (of the A-C-E, the blue line) from 34th Street-Penn Station to 53rd Street-5th Avenue. Fifth Avenue is one block west of Madison Avenue.

The 50th Street Station on the C and E trains is at 50th Street and 8th Avenue.Don't take the A, as it is an express train and will not stop at 50th Street.
