An opinion (one scenario):
If you're calling the prophesied "gathering of God's saints" the "rapture"... then, Jesus tells us that said gathering will occur "AFTER" the Great Tribulation:
"Immediately AFTER the Tribulation of those days shall the sun be darkened... and THEN shall appear the sign of the Son of Man in heaven... And He shall send His angels... and they shall gather together His elect..." (Matt.24:29-31).
The Bible, however, doesn't speak of an "imam"... 12th or any other number.
Daniel does write about "three" global powers extant on earth at the time of the end [of the Great Tribulation]... one of which he calls the "king of the south" who "pushes at" a "king of the north" [Dan.11:40]. And as these two factions clash with the king of the north responding to the "push" by invading the Middle East in a blitzkrieg-style attack ['...the king of the north shall come against him like a whirlwind...' - same verse] -- a third power approaches from the north.
Traditionally and historically... the "kings of the south" have been "Ptolemies" or "Egyptian."
Biblically speaking... while the religion of "Islam" wasn't even invented yet... the descendants of Ishmael made their presence felt throughout world history. And while an alleged "12th imam" is expected by many of Ishmael's descendants, today... Daniel's prophesied end time "king of the south" could serve to fulfill their expectations of the "12th imam" [Mahdi].
In these past few decades, the Islamic nations have been disjointed and ununited for the most part with no powerful central government to lead their aggressive life's mission of global dominance.
The recent upheaval in Egypt, however, may have begun to change that... and may lead to more and more Muslim nations overhauling and emerging with more aggressive and less "Israel-friendly, peace-process-friendly" Iranian-style governments.
Under the present global conditions: Bible prophecy would seem to point to a powerful unification of the Muslim nations under [possibly] an "Egyptian" leader... a "Ptolemy"... possibly the prophesied "king of the south."
If the defunct Egyptian government reemerges with a more aggressive, Iran-like outlook and attitude... the Islamic nations may begin to follow suit, fall into step -- and they may finally unite under a central leader, whom they may come to see as the "12th imam." [as global terror spreads, as the global economy weakens and finally crashes, as nations go bankrupt, collapse and fall].
If global events continue as they are... the three major world powers of the future likely to emerge from this growing global distress are Europe [the Beast, the 'king of the north'], the rioting Muslim nations [king of the south], and a combined Sino-Russian power "northeast" of Jerusalem.
"...tidings out of the east and out of the north shall trouble him..." (Dan.11:44).
Should this type of situation develop... this "king of the south" who may be viewed as "Mahdi" by Muslims the world over, will be a major player in the Great Tribulation... which, according to the Bible, occurs BEFORE Jesus returns and gathers His elect.
