Take the Uptown 1-2-3 (the red line), from 34th Street-Penn Station to 42nd Street-Times Square. Transfer at Times Square to the S shuttle train (color grey), or the Uptown 7 train (the purple line) if the shuttle isn't running. Take the S or 7 to 42nd Street-Grand Central (at Park/Lexington Avenues).

To take the subway from LaGuardia to Penn Station, you will need to first take the Q70 bus from the airport to the Roosevelt Ave-Jackson Heights subway station. From there, you can transfer to the E train, which will take you directly to Penn Station.

Since the eastern entrance to Penn Station is located on 7th Avenue, the distance is zero. However, the answer is deceiving because Penn Station is massive. It spans between 7th Avenue and 8th Avenue (the equivalent of 4 city streets), from east to west, and between 31st Street and 33rd Street from south to north. So, yes, it is true that the entrance to Penn Station is situated directly on 7th Ave. But, once inside, one might find himself walking several city blocks to get to the actual departure platform for his train or vice versa, from his train to 7th Ave.
