They did not agree to the Missouri Compromise. They were just out voted by the north.

The South allowed so much territory to be free states because they did not believe that there would be any more state made out of that territory because they thought of it as not settleable... so not many people would want to go there. Because there was a population requirement to request statehood, they thought that there would not be any more free states carved out of the "free" territory. (I know... a bit repetitious...) Southerners wanted to fight for their rights so they volunteered. They believed that slaves were their property and taking them away would be like taking away your house. Northerners were very against slavery.

Slavery in the South was it's economic backbone... and while most white people in the South didn't have slaves -- never the less-- most got some type of income from the institution of slavery. It is also important to remember: Slavery was a way of life for the Southern people and people do not normally like change, especially if someone else is trying to force it down their throat.

Beyond just Economics, the Confederacy had fewer people than the North. Of the 9 million southerners, only 5.5 million were white. Whereas the north had 22 million people. Four slave states, Delaware, Missouri, Kentucky and Maryland remained loyal to the union. Pockets of unionism existed elsewhere as well. Throughout the war, a steady stream of Blacks fled slavery to fight with the Union.
