US bills printed prior to the 1990 redesign had very few anti-counterfeiting features. Among other items they didn't have watermarks, security strips, color-shifting ink, or microprinting. About all you can look for would be:

Now a days there are security features on the 100 dollar bills to help you detect if the bill is counterfeit or not. One of the features is a watermark. If you hold the bill up to the light you will see a faint image of Benjamin Franklin to the right of the portrait.
When cashiers are handed a twenty dollar bill, they must mark it with red or black ink to check the authenticity. Shops use different methods to detect counterfeit paper money. One method is to mark it with a special pen, the ink from which will change colour if used on a counterfeit note. However it means a lot of marked notes are in circulation.

Vending machines use a bill acceptor equipped with sensors and a magnetic strip reader to authenticate and identify the denomination of the dollar bills. The sensors detect the unique patterns and features on each bill to determine its value and whether it is genuine or counterfeit. Once validated, the bill is stored securely until the transaction is completed.
