A test that you can do to confirm for example that your unknown is Bacillus subtilis from Bacillus megaterium is VP because B.subtilis is positive for VP and B.megaterium is negative for VP. There are plenty of test that you can do for example a gram stain, check if its aerobic, anaerobic, facultative anaerobic, check if its motile, is it a rod or cocci.. all those test help you find out more about your organism. There isn't only one test that you can do to confirm if your organism is from the Bacillus species you have to do multiple test because plenty of bacteria are gram positive, motile, rods, aerobic. Hope this helps:)
The Starch Hydrolysis, it form a white clearing zone around the bacteria

Is Bacillus subtilis motile or not?

Yes it is. The reason Bacillus is motile is because it is an aerobe. This means it thrives on oxygen. Inside the agar, oxygen is limited and usually it will move toward the surface where it is more oxygenated. Motility of Bacillus depends on the incubation time, environment and preparation of the agar. If all the other tests point towards Bacillus subtilis, you should be just fine.

What are the common biochemical tests used in identification of bateria?

The most common biochemical tests are gram stain, oxidase, catalase and coagulase tests. However, there are literally hundreds of biochemical tests that are commonly used to identify bacteria. For further information, check out MicrobeID.com, where you can find identification methods, keys, probabilistic databases, selective and differential media guides, as well as book reviews releated to bacterial identification. I would also recommend Bergey's Manual of Deterministic Bacteriology.

what are the biochemical tests for ascomycetes fungi?

Why is biochemical testing important?

A biochemical test is a test used mainly in microbiology with the main intention of detecting enzyme production. Biochemical tests can also, when used correctly, further narrow the search when looking at identifying an unknown microbe. The biochemical tests can narrow the search down to a specific genus and possibly even the species of the unknown microbe. In most cases when identification of an unknown microbe is needed, the microbe will usually be a bacterium.

What is the difference between bacillus coccus and spirillum?

Bacillus subtillus will test positive when performing a VP and Citrate test. Also, bacillus cereus is much more motile. It isn't as dependable as the chemical tests, but the cereus colonies will appear wispy and more widespread on an agar plate than those of subtilis.
