You could also use 知らない (Shiranai), which is 'I don't know'.
If you mean to say you don't understand Japanese, then you would use 日本語を話しません (Nihongo wo hanashimasen), which is 'I don't speak Japanese'. Note that the 'w' in 'wo' is silent, so you would say it as 'o'.
You wouldn't use desu with the last one, because 'masen' replaces it.
