1. Add approximately 50 mL of distilled water to a

100ml volumetric flask on a magnetic stirrer.

2. Add and dissolve 1.4705gm of Sodium citrate.

3. Then with the help of Citric Acid maintain the pH of the solution upto 4.5.

You have to simply dissolve streptozotocin (STZ) in the freshly prepared citrate buffer pH 4.5. But first you have to calculate the amount of STZ and the volume of citrate buffer required. Here's an example for a rat weighing 150 gms. STZgiven at 45 mg/kg of body weight. So for 150 gms you need 6.75 mg of STZ. Now you need to inject this in the abdominal cavity of rat so prefer to use minimum volume of buffer. So here, we'll dissolve 9 mg of STZ in 1 ml of Citrate buffer pH 4.5 and inject 0.75 ml of this solution into rat. Citrate buffer as in British Pharmacopeia It would be better if STZ is dissolved just before use.

To make one liter of buffer, weigh out point one mole of citric acid. Dissolve it in about six hundred milliliters of water. Adjust the pH with six M NaOH until the pH is four point five. Bring the volume of the solution to one liter with water.

To make a citrate buffer Mix 21 g citric acid in 1 L distilled water, and 29.4 g sodium citrate in 1 L distilled water. If you only have citric acid, mix 2.1 g in just under 1 L distilled water. You can adjust the PH level by adding 1M sodium hydroxide while gently stirring the solution using a magnetic stirrer.

How does magnesium citrate work?

Magnesium is the pure element, however it is not found in nature by itself, it always bonds with another element. So you have many forms of magnesium such as magnesium Aspartate, bicarbonate, Carbonate, Chloride, Citrate, Gluconate, Glycinate, Hydroxide, Lactate, Malate, Orotate, Oxide, Phophate, Picolinate, Propionate, Stearate, Sulfate and Taurate, to name a few. All these different forms have one other element that they are bonded with magnesium + citric acid = magnesium citrate for instance. These different forms of magnesium have two specific qualities A. they have percentage of actual elemental magnesium i.e. how much of the combination is actual pure magnesium e.g. magnesium carbonate is 45% actual magnesium, where as magnesium sulfate is only 10% actual magnesium B. they have a rough percentage of bioavailability (how absorbable and thus useable they are for the body). For instance magnesium citrate is 90% bioavailable and so most of it could be potentially used and absorbed by the body. Magnesium oxide for instance is only 4% bioavailable for the body and so if one took magnesium oxide which had 100mg of actual magnesium in it, there would only be 4mg that the body could possible use.
