When isolating DNA from blood, white blood cells (WBC's) are the target. This is because RBC's do not contain a nucleus and therefore do not contain DNA.
The function of the lysis buffer is to help in the lysis (or breaking) of white blood cells. WBC's must first be lysed so that the DNA may be released from inside the cell.
