Queen Elizabeth I would be a distant cousin of Queen Elizabeth II. Both Elizabeths are descentants of Henry VII.

Elizabeth I was the granddaughter of Henry VII through her father Henry VIII. Elizabeth II is a direct descendant of his through his daughter Margaret, Queen of Scotland and her great-grandson James I-who was the great-grandfather of George I-who is the 9th great grandfather of Elizabeth II

The mother of Elizabeth II, aka Lady Elizabeth Bowes-Lyon aka the Queen Mum aka the character played by Helena Bonham-Carter in The King's Speech, was the wife of a (reigning) king. As such, she doesn't get an ordinal. During her husband's reign, she was Queen Elizabeth, but once he died, she became the Queen Dowager. Elizabeth I (and Elizabeth II) are both regnant queens--they were both on the throne themself, rather than being married to the ruler.
