Most likely the most famous battle of the war was Gettysburg, PA, from July 1-3, 1863. There, the USA under George Meade's Army of the Potomac decisively defeated Lee's Army of Northern Virginia. The significance of the victory was that the USA had shattered Lee's dreams to force a victory on Union soil, and had set a precedent of the future victories to come. The war would be won almost two years later, but if the United States had lost Gettysburg, it was very likely that the Confederacy would have won the war, keeping all of its slaves.
