Careful, you can't call it a "blockade" that would be an act of war and would have justified the USSR issuing a declaration of war on the US. Kennedy only called it a "quarantine". (even if effect was same)
It was a hugely symbolic gesture, but more or less ineffective despite movie portrayals. The key weapons were already in Cuba and ready to be deployed, and the blockade wouldn't have done anything to affect that reality.
In the end, it was a back-door negotiation by JFK representatives through unofficial channels that ended the stalemate. JFK offered to remove obsolete Jupiter missiles from Turkey in exchange for the removal of the nukes in Cuba. The missiles in Turkey were the real catalyst for the Cuban missiles anyway.

Times were tense during the Cuban Missile Crisis to say the least; there were already several instances between U.S. and Soviet forces that could've led to a further escalation. A Naval Blockade is considered an act of war. To avoid that, the Kennedy Administration used the term "Naval Quarantine" (semantics), as the crisis played out also on the world stage in the United Nations. Moderates on both sides were doing everything possible to prevent the crisis from escalating into a full blown nuclear war.

The Soviet Union began placing nuclear armed guided missiles in Cuba, where they could be used to attack to US with almost no warning. The US declared that any attack coming from Cuba would be considered an attack from the Soviet Union, would result in retaliation to the Soviet Union, and directed that the missiles be removed from Cuba. The US Navy began a shipping blockade and stopped additional missiles from being taken to Cuba.
